چکیده

Studies on the nucleolytic activity of cell homogenates of Tetrahymena pyriformis indicated that enzyme activity was, among other factors, a function of the age of the culture (1, 2). It was shown that the specific activities of ribonuclease (RNase) (EC 2.7.7.h) and acid phosphatase (APase) (EC 3.1.3.2) in crude homogenates prepared from stationary-phase cultures were consistently higher than those from log phase by a factor of at least two (2). However, in contrast to these two enzymes, the level of a potassium-activated ribosomal phosphodiesterase (PDase) (EC 3.1.4.1) was reported to be maximal at the end of exponential multiplication (3). Our results on the level of APase in T. pyriformis GL, however, could not be reconciled with the data of Klamer and Fennel (1), who reported increased APase activity in strain W only during the exponential death phase. Therefore, it was of interest to further investigate the activity of these nucleases in more detail. In addition, we wish to report here the effect of puromycin, an inhibitor of protein synthesis, on the change in activity of RNase and APase during growth.

Secretion of Acid Hydrolases and Its Intracellular Source in Tetrahymena Pyriformis

Axenic Tetrahymena pyriformis, syngen 1, mating type II cells were grown in Cox's defined medium. When washed and transferred into nonnutrient dilute salt solution or resuspended in the defined medium, the intact cells secrete acid hydrolases into the medium. Cells starving in the salt solution release in 5 hr about two-thirds of their beta-glucosidase, beta-N-acetylglucosaminidase, alpha-gluco...

Effect of different toxic compounds on ATP content and acid phosphatase activity in axenic cultures of Tetrahymena pyriformis.

The sensitivity of protozoa, and particularly ciliated protozoa, to environmental changes suggested a study on the physiological responses arising from exposure to toxic compounds. Tetrahymena pyriformis was used as a test organism in a set of miniaturized assays. The physiological response of this ciliate was assessed in terms of adenosine-5'-triphosphate content and acid phosphatase activity ...

Micronuclear Ribonucleic Acid in Tetrahymena Pyriformis

The presence of RNA in the micronucleus of Tetrahymena pyriformis was detected by electron microscope radioautography after incubation with tritiated precursors. The specificity of RNA labeling was shown by ribonuclease digestion. The period of appearance of labeled RNA in the micronucleus is approximately coincident with the DNA synthesis period for the micronucleus. Pulse-chase experiments sh...

The Fate of Mitochondria during Aging in Tetrahymena Pyriformis

During the growth cycle of Tetrahymena pyriformis the mitochondria undergo changes in position, number, and structure. Ciliates in the logarithmic growth phase possess elongated mitochondria which are aligned along the plasma membrane and are closely associated with the kinetosomes and kinetodesmata. Mitochondria appear to divide across the long axis at this time, resulting in two or more produ...

Relationship of adenosine 3',5'-monophosphate to growth and metabolism of Tetrahymena pyriformis.

The concentration of adenosine 3',5'-monophosphate (cyclic AMP) and the activity of adenylate cyclase were determined for the first time in conjuncation with cyclic 3',5'-nucleotide phosphodiesterase (phosphodiesterase) during the growth cycle of Tetrahymena pyriformis. High levels of cyclic AMP observed during early exponential and late stationary phases were associated with elevated adenylate...
